Summary Statement: ADARC Program Project on HIV/SIV Vaccine Development
PAGE 34 1 P01 AI43042-01 9 SEP Ho, Dav id D. 4'f 4C CCSf while the construction of an env protein that stimulates antibodies to critical epitopes is a worthwhile objective, it has already been pursued by many other investigators. In addition, it is completely unclear which of the participating investigators would be doing what, thereby making it nearly impossible to judge the ability to make the necessary constructions.; RESOURCES AND ENVIRONMENT: The facilities at both ADARC nd PHRI, n terms of both space and equipment, are more than adequate to perfor roposed experiments. Most laboratories (Project 1) are located on two floors of one facility and a closely integrated primate research facility. The general quality of the facilities is high, and both institutions are well positioned to conduct hjgh quality, interactive research. The full commitment of the ADARC towards AIDS research is unquestionable, and collaboration between the investigators of all projects andcores is excellent. PROGRAM ADMINISTRATION: The leadership and research experience of the Principle Investigator, Dr. D. Ho and his team are outstanding. Dr. Ho is known worldwide for his work on HIV. The team that he has assembled at the Aaron Diamond AIDS Research Center including Drs. Moore, Connor, Nixon, and Marx, have also contributed greatly to our understanding of HIV. The staff, for the most part, have the necessary experience to conduct the work. OVERALL IRG ACTION: Based upon the evaluation by the IRG of the scientific and technical merit of the projects and cores recommended for support, and the interactions in the resulting overall program consisting of Projects 1, 2, and 3, and Cores B, and C, this application is, scored in the good range range with a numerical value of 330. PROJECT j: Live, Attenuated SIV Vaccine and Correlates of Protection Critique: The comments below represent essentially unedited comments from the reviewers of this application. They are included to indicate the range of comments made during the discussion, and may not reflect the final thinking of the committee. The RESUME and OVERALL CRITIQUE sections above summarize the final opinion of the committee after the discussion. REVIEHER A: This 5-year project is planned to further analyze a previous study and perform a second study using 14 macaques to prospectively evaluateI SIV-specific CTL and other cellular immune responses. It is very likely that many of these studies are well underway or have already been finished based on data given in the application or at meetings. In addition, similar 89.6 SHIV re-challenge and mu nge o pro ec e - f es have b ne in other research laboratories although the SIV-GFP iso te was used. It is very surprising that despite the collection of LN for viral RNA levels and cytokine expression, very little is to be done with them. says are performed on PBMConl-. - / Continued
