Summary Statement: ADARC Program Project on HIV/SIV Vaccine Development
PAGE 50 1 P01 AI43042-01 25 SEP Ho, David D. In summary, the strengths of the Primate Core include: (1) the excellent research primate facility; (2) an exceptional expertise of the Principal Investigator in non-human primate retrovirology; and (3) the well established macaque models with SIV/SHIV infections. The impact on the achievement of the stated objectives is rated as outstanding. With regard to the budget, the costs for purchase of 170 new macaques for the three projects, their per diem expenses, clinical studies, laboratory tests, surgical biopsies, and necropsies, are not specifically described in the primate core. In addition, the cost of some items, such as the total number of animals per diem, blood draws, and clinical laboratories are not clear in years 02-05 f the 5-year budget summary. Because of interactions (e.g., the shared use of macaques, blood samples, or tissue specimens) and certain laboratory supports (e.g., tests or assays) between the three projects and central cores, there may be an overlap in the proposed budget. REVIENER C: The primate animal core will provide support to investigators from three projects in doing SIV/SHIV studies in rhesus macaques. Operation of the facility, which is located in Tuexedo, NY, is being negotiated with ADARC which has an active primate laboratory at this center since 1993. A full-time staff consisting of animal research technicians and two veterinarians provides animal care. Experimental protocols for the primates are reviewed monthly, and the Institutional Animal Care and Use Committee inspects the facility three times a year. The laboratory is prepared to house singly-caged macaques under biosafety level "2+" conditions. The staff members which will be responsible for veterinary care and experimental procedures are experienced and highly skilled in various procedures in primates. Work outlined by three different investigators involving SIV infection of rhesus macaques over the proposed 5-year period will require purchase of 170 animals. A review of animal requirements for the proposed studies indicates that the animal requests are justified, and that the facility is adequately prepared to assist with these projects. A review of proposed primates to be purchased during the proposed 5-year period of the project indicates that the projected expenses are moderate and appropriate for the work outlined. Human Subjects: code 10 Nomen and Minorities Representation: not applicable Animal Welfare: code 30 Budget: The proposed budget is recommended for approval as requested.
