Summary Statement: ADARC Program Project on HIV/SIV Vaccine Development
PAGE 48 1 P01 AI43042-01 23 SEP Ho, David D. The standard assays for monitoring the immune responses elicited by the proposed vaccines will be carried out according to published procedures. Characterization of humoral and cellular responses will be performed and parameters, such as the measurement of cytokine production, will be quantitated. Molecular analyses, such as TCR repertoire analysis and MHC typing, will be available. The laboratories are well equipped to conduct the proposed assays and the personnel is qualified to provide the Core services without any serious limitations. The requestedtbudget for the Immunology Core is appropriate. REVIEWER C: This core is able to meet its objectives of quan-titation of specific humoral responses (neutralization assays, antibodyfinding, and titration), lymphocyte proliferative responses, CTL responses, TCR repertoire analysis, MHC typing, ADCC, CD8+ suppressor activity, chemokine assays, macrophage and dendritic cell generation and characterization, and immunochemical staining. As written, this core is critical to the objectives of this P01 application. Human Subjects: code 10 Women and Minorities Representation: not applicable Animal Welfare: code 30 Budget: The proposed budget is recommended or approval as requested. IRG Action: This core was rated a appropriate. CORE C: Primate Core (Dr. P. Marx) Critique: The comments below represent essentially unedited comments from the reviewers of this application. They are included to indicate the range of comments made during the discussion, and may not reflect the final thinking of the committee. The RESUME and OVERALL CRITIQUE sections above summarize the final opinion of the committee after the discussion. REVIEWER A: The Primate Core will provide in-depth support for all three projects of this application involving SIV/SHIV studies in macaques. The staff of the core is extremely experienced in providing support and resources for infectious disease research in primates, and is entirely appropriate for conducting this work. The Aaron Diamond AIDS Research Center has a long-standing relationship with the Laboratory for Experimental Medicine and Surgery in Primates (LEMSIP), and has maintained a laboratory and offices there for a number of years.
