We anticipate no additional purchases of equipment during the project period. Supplies and Other Expense At Davis, a total of 560 male, adult, CD-1 mice will be used in this study. In the first year, 140 will be exposed to the amyl nitrite vapor at 330 ppm for six months, as described by Al-Bayati and Raabe, 1992, and 140 will be used as a control. In the second year, 140 mice will be exposed to amyl nitrite and clean air as described above, but at a lower or higher concentration or for a longer exposure time depending upon the result of the first exposure. The animals will be observed daily and clinical signs will be recorded and evaluated. Animals will be weighed twice a month. Five to ten exposed and five to ten control animals will be sacrificed after one to two weeks. Thymus, lymph nodes, spleen, liver, kidney, lung and brain will be taken for gross and microscopical examination as described by Al-Bayati, et. al, 1989. Blood and serum samples will be taken from each animal for standard hematology and serum chemistry analysis as described by Al-Bayati, et. al, 1991. Funds requested in Year One for non-animal costs are based on actual historical consumption of these items. Funds for animals and animal care are based on rates at UC Davis. Figures for years two through three reflect a 4% adjustment for anticipated price increases. Travel Funds are requested for two round trips per month between UC Berkeley and UC Davis, to be undertaken by the Berkeley research personnel, at 136 miles per round trip, reimbursable at $.24 per mile. These trips will be for the purpose of co-examination of the mice exposed to nitrite inhalants, and participation in evaluation of the progress of the toxicity studies.
