CCR5 Ligands in HIV Vaccines
cryopreserved samples versus fresh ones; b) protocol and length of cryopreservation; c) time elapsed from immunization. Aim 3: To study antigen specific release of CCR5 ligands in samples obtained from vaccine trials in humans. These studies will clearly establish the value and utility of our proposed approach. The studies in this aim will be carried out using samples from the HVTN (from subjects immunized with canarypox-based immunogens). Chemokine responses will be established and compared with standard analyses such as antigeninduced proliferative responses, neutralizing antibodies and cytotoxic T cells analyses. We expect that the accomplishment of these aims will allow for a complete evaluation of immune responses to vaccines against HIV infection, by potentially contributing the long sought correlate of protection. NB: AIM 3 is to be performed in collaboration with the HVTN (pending approval) and other vaccine networks. In order to perform all of the proposed assays, about 25 x 106 cells/subject are required. See below for specific information on how many cells are required for each assay. Ideally we would like to receive samples obtained from subjects immunized with Canarypox-vectored immunogens. See Below for information on samples needed for the proposed studies.
