CRIXIVAN - CLINICAL TRIAL UPDATE

PROTOCOL 067

CRIXIVANTM* in combination with efavirenz
101 nRTI-experienced patients randomised to CRIXIVAN
(800/1000mg q8h) plus efavirenz (600mg once daily), or
CRIXIVAN alone. Monotherapy arm discontinued at week 12;
patients offered option of adding efavirenz and stavudine
(D4T) to align with current treatment standards. Mean HIV
RNA = 5.06 log0lo, mean CD4 283 cells/mm3.
* 60-week data presented January 1998 at the 5th Conference
on Retroviruses and Opportunistic Infections, Illinois, USA
showed 89 percent of patients on the CRIXIVAN/efavirenz
combination had undetectable viral load (<400 copies/mL).
* 72-week data to be presented at the 12th World AIDS
Conference, Geneva, Switzerland in June 1998
CRIXIVAN twice-daily in combination with efavirenz
* 71 PI- and NNRTI-naive patients randomised to either threetimes-a-day therapy with CRIXIVAN (1000mg q8h) plus
efavirenz (600mg qhs or a twice-daily regimen of CRIXIVAN
(1200mg q12h) plus efavirenz (300mg q12h)
* 16-week follow-up to be presented at the 12th World AIDS
Conference, Geneva, Switzerland in June 1998 suggests that
the safety profile and antiviral activity of twice-daily dosing are
comparable to current standard of CRIXIVAN three-times-aday dosing.
