associated with growth of human T cells, a suppressive response of the SCID recipient, or a general shortage of human macrophage function. HIV infection of hu-PBL-SCID mice A major incentive in the development of the hu-PBL-SCID mouse was to provide a small animal model for AIDS research. We have now completed studies [5] showing that hu-PBL-SCID mice can be infected either with cell-free human immunodeficiency virus (HIV) or with HIV-infected syngeneic T lymphoblasts. Virus can be cultured from the spleen, peripheral blood, and lymph nodes of infected mice, and in situ hybridization confirms infection of human cells in the SCID spleen. In addition, HIV-specific sequences can be amplified from hu-PBL-SCID spleen by the polymerase chain reaction. Figure 3 shows the results of an experiment in which hu-PBL-SCID mice were infected with either cell-free or cell-associated LAV-1/Bru strain HIV-1. Sixteen identical hu-PBL-SCID mice were constructed by the injection intraperitoneally of 40 x 106 PBL from a single EBV-seronegative donor.
